3 Carcinogenicity

چکیده
The objective genotoxicity assay the methodology of the carcinogenicity and working group was to assess available for predicting and identifying the human carcinogenicity of chemicals that are subject to review and testing under the Toxic Substances Control Act (TSCA), with emphasis on existing chemicals. Specific objectives were to 1) identify existing assays for assessing or predicting the human carcinogenicity of chemicals, 2) indicate which assays are the most reliable for the prediction of human carcinogenicity, and 3) assess the reliability of low cost predictive assays. This mandate included an assessment of genotoxicity assays in the context of their predictive value for carcinogenicity, but did not include an assessment of other health implications of genotoxicity. The objectives are summarized in table 3-1.

Alternative carcinogenicity screening assay using colon cancer stem cells: A quantitative PCR (qPCR)-based prediction system for colon carcinogenesis.
The carcinogenicity of chemicalsin the environment is a major concern. Recently, numerous studies have attempted to develop methods for predicting carcinogenicity including rodent and cell-based approaches. However, rodent carcinogenicity tests for evaluating the carcinogenic potential of a chemical to humans are time-consuming and costly. This study focused on the development of an alternative...
متن کاملAnimal carcinogenicity studies: 1. Poor human predictivity.
The regulation of human exposure to potentially carcinogenic chemicals constitutes society's most important use of animal carcinogenicity data. Environmental contaminants of greatest concern within the USA are listed in the Environmental Protection Agency's (EPA's) Integrated Risk Information System (IRIS) chemicals database. However, of the 160 IRIS chemicals lacking even limited human exposur...
متن کاملGenotoxicity, carcinogenicity, and mode of action of the fried food mutagen 2-amino-3-methylimidazo[4,5-f]quinoline (IQ).
Because mutagens typified by 2-amino-3-methylimidazo[4,5-f]quinoline (IQ) observed in cooked foods are widely consumed, detailed studies of their biochemical and biological properties including carcinogenicity are most important. IQ induces unscheduled DNA synthesis in liver cells, which when taken together with its powerful mutagenicity in the Salmonella typhimurium test system, predicts carci...
متن کاملAtrazine Citation for Most Recent Iarc Review
Current evaluation Conclusions from the previous Monograph: Atrazine is not classifiable as to its carcinogenicity to humans (Group 3). There is inadequate evidence in humans for the carcinogenicity of atrazine. There is sufficient evidence in experimental animals for the carcinogenicity of atrazine. The Working Group concluded that the animal mammary tumors associated with exposure to atrazine...
متن کاملImplications of the lack of accuracy of the lifetime rodent bioassay for predicting human carcinogenicity.
The NTP lifetime rodent bioassay (LRB) is the "gold standard" for predicting human carcinogenicity. Unfortunately, little attempt has been made to validate it against human carcinogenicity. Here we show that the extremely limited data available do not support either of the two common interpretations of LRB results. If a risk-avoidance interpretation is used where any positive result in a sex/sp...
